3つの要点

✔️AIとシステム生物学を利用して、神経外傷用の神経保護薬に使う物質の種類を調べた。

✔️最良の組み合わせの物質(NeuroHealと呼ぶ)は、既存の薬物とは異なり、かつ抗炎症や運動機能回復などの効果を発揮することがわかった。

✔️研究の結果は、NeuroHealの有効性を示すとともに、創薬に対してネットワーク中心のアプローチが十分な価値をもつということを実証するものでもあった。

概説

末梢神経根剥離の治療のための神経保護薬を同定するために、我々はシステム生物学的アプローチと人工知能(Artificial Intelligence;AI)を使用した。

根剥離後の運動ニューロンで発生する神経変性と神経保護プロセスについて学習した知識に基づいて、我々はタンパク質ネットワークを構築し、それらを数学モデルに変換した。

我々の前臨床モデルからの不偏なプロテオミクスデータは、機械学習アルゴリズムおよび数学的解法に課されるべき制限のために使用された。

解法は、潜在的な神経保護薬として再利用された薬の組み合わせを同定することができ、それらを前臨床モデルで検証した。

神経保護された運動ニューロンで最良のもの──NeuroHeal──は、抗炎症特性を発揮し、そして機能的な運動回復を促進した。

NeuroHealは、神経保護効果に不可欠なサーチュイン1の活性化を促した。これらの結果は、創薬におけるネットワーク中心アプローチの価値を裏付け、神経系外傷に対する外科的修復を伴う補助療法としてNeuroHealが有効であることを実証している。
